3. In order to prevent the liquid level from dropping after the ice bath melts and causing no load, the sample should be placed in a 1.5ml EP tube. Remember to fix the EP tube.
4. Ultrasonic parameter setting of ultrasonic pulverizer: set the working parameters of the instrument. For samples that are sensitive to temperature (such as bacteria), ice bath is generally used outside. The actual temperature must be lower than 25 degrees, and protein nucleic acid will definitely not be denatured. . 
A. Time: The ultrasound time should not exceed 5 seconds each time, and the gap time should be greater than or equal to the ultrasound time to facilitate heat dissipation. The time setting should be based on the principle of short ultrasound time and multiple ultrasound times, which can extend the life of the ultrasound machine and probe. 
B. Ultrasonic power of the ultrasonic pulverizer: it should not be too large to avoid splashing or foaming of the sample. If the sample capacity is less than 10ml, the power should be within 200W.
